بررسی الگوی بیان ژن های کیتیناز و بتا-1و3 گلوکاناز در گیاه نخود آلوده به بیماری برق‌زدگی

چکیده

القای مقاومت نسبت به پاتوژن‌ها از راهکارهایی است که گیاهان برای مقابله با تنش‌های زیستی به‌کار می‌برند. آنزیم‌های کیتیناز و بتا-1و3 گلوکاناز جزء پروتئین‌های قابل‌القاء هستند که در پاسخ به عوامل بیماری‌زا توسط گیاه، سنتز می‌شوند. در این تحقیق، بیان ژن ایزوفرم‌های اسیدی و بازی این دو آنزیم در سطح مولکولی در گیاه نخود زراعی مورد مطالعه قرار گرفت. به‌این‌ترتیب که سطح تظاهر ژن‌های مذکور در نمونه‌های تیمارشده و شاهد، در ژنوتیپ نخود حساسMCC403 و ژنوتیپ مقاوم MCC496 نسبت به آلودگی با نژاد شِش قارچ برق‌زدگی (Ascochyta rabiei) در 0،6، 12، 24، 48 و 72ساعت بعد از آلودگی با استفاده ازروش ارزیابی نیمه کمیRT-PCR بررسی شد. نتایج، حاکی از افزایش بیان ایزوفرم اسیدی هر دو ژن کیتیناز و بتا-1و3 گلوکاناز پس از آلودگی بود. هردو ژن مذکور در 24ساعت اولیه پس از تلقیح، حداکثر بیان را در ژنوتیپ مقاوم نشان دادند. این در حالی است که در ژنوتیپ حساس در ساعات اولیه بعد از آلودگی، تظاهر دو ایزوفرم اسیدی به حداکثر می‌رسد و تا 72ساعت بعد از آلودگی، سطح بیان در هردو ژنوتیپ به کمترین سطح تظاهر می‌رسد. ایزوفرم بازی کیتیناز در ژنوتیپ حساس به‌میزان بیشتری بیان شد و ایزوفرم بازی
بتا-1و3گلوکاناز تظاهر پیدا نکرد. در کل، می‌توان نتیجه گرفت که احتمالاً ایزوفرم‌های اسیدی ژن‌های کیتیناز و گلوکاناز در اعطای مقاومت در برابر بیماری برق‌زدگی نخود نقش دارند.

چکیده [English]

Induced resistance to pathogen is one of the ways which plants use against biotic stresses. Pathogenesis-related proteins(PR), including chitinase and β-1,3-glucanase synthesized by the plant in response to pathogenic infection. In this investigation, the expression of two isoforms: acidic and basic of chitinase and β-1,3-glucanasegenes were analyzed at 0, 6, 12, 24, 48 and 72 hours after fungal inoculation of chickpea genotypes (MCC496 as resistant and MCC403 as sensitive) with race 6 of Ascochyta rabiei by semi-quantitative RT-PCR. The results showed that the expression of both acidic isoforms of chitinase and β-1,3-glucanase genes were increased at 24 and 48 hours, respectively after fungal inoculation in genotype MCC496, but the expression level in sensitive genotype increased in early time after inoculation and then decreased. The basic isoform of chitinase was expressed in sensitive genotype more than the resistant genotype. The basic isoform of β-1,3-glucanase was not expressed in these genotypes. Totally, acidic isoforms of these enzymes specifically chitinase would play roles in plant defense against Ascochyta blight
