اثر تاریخ کاشت، فاصله ردیف و مقادیر بذر بر عملکرد دانه و پروتئین گیاه باقلا (Vicia faba L.) در شهرستان رشت
بهمنظور مطالعه اثر تاریخ کاشت، فاصله کشت و مقدار بذر بر عملکرد دانه و عملکرد پروتئین باقلا، آزمایشی بهصورت اسپلیت فاکتوریل در قالب طرح بلوک های کامل تصادفی در سه تکرار طی دو سال زراعی 89-1387 در ایستگاه تحقیقاتی مؤسسه تحقیقات برنج کشور در رشت اجرا گردید. در این آزمایش سه تاریخ کاشت 10 و 25مهر و 10آبان بهعنوان کرت های اصلی و سه فاصله بین ردیف های کشت 30، 40 و50سانتی متر و دو میزان بذر100 و150کیلوگرم در هکتار بهعنوان کرت های فرعی منظور شدند. نتایج مقایسه میانگین نشان داد که سال اول، از نظر اغلب صفات مورد بررسی نسبت به سال دوم، از برتری قابلملاحظهای برخوردار بود. بین تاریخ های کاشت، تاریخ کاشت 25 و 10مهر، بیشترین وزن100دانه، تعداد شاخه جانبی، تعداد غلاف در بوته و عملکرد دانه و پروتئین را بهخود اختصاص داد و بهطور مشترک در یک گروه قرار داشتند. بین فواصل کشت، فاصله کاشت 40 و 30سانتیمتر و بین مقادیر بذر، میزان بذر150کیلوگرم در هکتار، بیشترین عملکرد دانه و پروتئین را بهخود اختصاص دادند. تاریخ کاشت 10مهر با میانگین 234روز بیشترین طول دوره رشد را دارا بود. نتایج حاصل از تحقیق، حاکی از آن بود که کاشت باقلا در تاریخ کاشت 10 و 25مهر ماه، فواصل کشت 30 و 40سانتی متر و مقدار بذر 150کیلوگرم در هکتار بهدلیل بیشتر بودن عملکرد دانه و پروتئین، در اراضی شهرستان رشت قابل توصیه میباشد.

بررسی امکان کاشت پاییزه نخود در شرایط آب و هوایی سراوان
در برخی مناطق گرم و خشک، وجود فصل رشد مناسب در طول پاییز و زمستان و وقوع درجهحرارت بالا در بهار سبب گرایش به کشت نخود در پاییز شده است. بنابراین بهمنظور بررسی مقدماتی امکان کاشت پاییزه نخود
(Cicer arietinum L.) در شرایط آب و هوایی سراوان، آزمایشی در پاییز سال1388 در مزرعه تحقیقاتی دانشکده کشاورزی و منابع طبیعی سراوان بهصورت کرتهای خُردشده در قالب طرح بلوک های کامل تصادفی با سه تکرار اجرا شد. کرت های اصلی شامل سه تاریخ کاشت (20مهر، 11آبان و اول آذر) و کرت های فرعی شامل پنج ژنوتیپ نخود MCC488, MCC208, MCC258, MCC770, MCC361)) بودند. تاریخ کاشت تأثیر معنیداری بر عملکرد دانه، ارتفاع بوته، تعداد غلاف در بوته، تعداد دانه در بوته، تعداد غلاف خالی، وزن100دانه، عملکرد زیستی، عملکرد اقتصادی و شاخص برداشت داشت و گیاهان کشتشده در 20مهر بیشترین عملکرد دانه را داشتند. اثر ژنوتیپ نیز بر کلیه صفات اندازهگیریشده معنی دار شد و ژنوتیپ MCC361 با میانگین عملکرد دانه 6/204گرم در مترمربع بیشترین عملکرد را دارا بود. اثر متقابل تاریخ کاشت و ژنوتیپ، معنیدار شد و بیشترین عملکرد دانه را ژنوتیپMCC361 در تاریخ کاشت اول (20مهر) با میانگین 255گرم در مترمربع و کمترین آن را ژنوتیپMCC770 در تاریخ کاشت سوم (اول آذر) با میانگین 55/17گرم در مترمربع داشتند. همبستگی مثبت و معنیداری بین عملکرد دانه با تعداد غلاف در بوته (**84/0r= )، تعداد دانه در بوته (**63/0r= )، عملکرد زیستی (**55/0r= ) و شاخص برداشت (**55/0r= ) مشاهده شد. نتایج این مطالعه نشان داد که هرچند کاشت پاییزه نخود در منطقه سراوان موفقیتآمیز بوده است، ولی برای حصول نتایج مطمئنتر، تداوم اینگونه آزمایشها و بررسی دقیقتر واکنش ارقام نخود به کاشتهای پاییزه توصیه میشود.

بررسی مزیّت نسبی طرح آلفا لاتیس نسبت به طرح بلوکهای کامل تصادفی در آزمایشات مقایسه عملکرد نخود زراعی (Cicer arietinum L.)
ساختار طرحهای آلفا لاتیس این امکان را فراهم میسازد که اثر بلوک ناقص از اثر باقیماندۀ کرتها حذف شده و دقت مقایسات بین ژنوتیپها در یک بلوک ناقص به حداکثر برسد. در این تحقیق، بهمنظور ارزیابی ژرمپلاسم بینالمللی نخود از لحاظ عملکرد دانه و سایر صفات زراعی و مقایسه طرحهای آلفا لاتیس با طرح بلوکهای کامل تصادفی از نقطهنظر توانایی شناسایی لاینهای مطلوب، دو آزمایش خزانة بینالمللی لاینهای برگزیدۀ نخود
(Chickpea International Elite Nursery) با عناوین CIEN-S-2010 و CIEN-LA-2010 در سال زراعی
90-1389 در شرایط دیم ایستگاه سارال کردستان به مرحله اجرا درآمدند. این دو آزمایش هرکدام شامل 45لاین آزمایشی نخود کابلی بودند که بهصورت آلفا لاتیس 5×9 با دو تکرار، در نیمه دوم اسفند1389 کشت گردیدند. با توجه به ویژگیهای آزمایشات، عملیات اجرایی و یادداشتبرداریهایی مانند تعداد روز از کاشت تا گلدهی، تعداد روز از کاشت تا رسیدگی، ارتفاع بوته، وزن100دانه و عملکرد دانه در موعد مقرر انجام شدند. بر اساس تجزیه واریانس، در هر دو آزمایش، اختلاف معنیدار بین ژنوتیپهای تحت مطالعه وجود داشت. از خطای معیار میانگین عملکرد ژنوتیپها برای برآورد مزیت نسبی طرح آلفا لاتیس استفاده شد. نتایج نشان داد که تجزیه آلفا لاتیس، کارآیی بیشتری نسبت به بلوکهای کامل تصادفی در هر دو آزمایش داشته و موجب تغییر در رتبة عملکرد دانه در لاینهای آزمایشی شد. در مجموع، لاینهای FLIP 05-55C (با عملکرد 992کیلوگرم در هکتار) و FLIP06-159C (با عملکرد 773کیلوگرم در هکتار) بهترتیب در آزمایشات CIEN-LA-2010 و CIEN-S-2010 رتبه اول را بهدست آوردند. از نتایج این تحقیق در مطالعات آتی میتوان استفاده کرد.

بهمنظور ارزیابی تأثیر نژادهای ریزوبیومی و ریزوباکتریهای محرک رشد گیاه (PGPR) بر تخصیص مواد فتوسنتزی و برخی پارامترهای مرتبط با رشد گیاه نخود زراعی (Cicer arietinum L.) رقم آرمان، آزمایشی بهصورت کرتهای خُردشده در قالب بلوکهای کامل تصادفی در مزرعه پژوهشی دانشگاه زنجان به مرحله اجرا درآمد. در این آزمایش، سطوح آبیاری در دو سطح (آبیاری مطلوب در طی فصل رشد و عدم آبیاری در کل دوره رشد) در کرتهای اصلی و سطوح کودی در 7 سطح (شاهد یا عدم مصرف کود شیمیایی و بیولوژیکی، مصرف 50کیلوگرم اوره در موقع کاشت، تلقیح بذر با Mesorhizobium ciceri نژادSWRI-3، تلقیح بذر با Mesorhizobium ciceri نژادSWRI-17، تلقیح بذر با PGPR، تلقیح مشترک با نژادهای ریزوبیومی (SWRI-3+SWRI-17) و تلقیح مشترک با نژادهای (SWRI-3+SWRI-17+PGPR) در کرتهای فرعی قرار گرفتند. نتایج این بررسی نشان داد که تیمارهای SWRI-17 و SWRI-3+SWRI-17+PGPR)) در شرایط تنش خشکی بهدلیل تخصیص بیشتر مواد فتوسنتزی به نیامها و تخصیص کمتر مواد فتوسنتزی به ساقهها و برگها، عملکرد دانه بیشتری تولید کردند. در شرایط آبیاری مطلوب نیز تیمار SWRI-3+SWRI-17+PGPR)) با تخصیص بیشتر مواد فتوسنتزی به برگها که احتمالاً باعث جذب بیشتر تشعشع میشد، عملکرد دانه بیشتری تولید کردند. بر اساس نتایج این بررسی، تلقیح بذر با ترکیبی از کودهای زیستی ریزوبیومی و PGPR در هر دو شرایط آبیاری مطلوب و دیم، باعث حصول عملکرد بیشتر نسبت به شاهد کودی یا مصرف کود نیتروژنه اوره میشود.

کارآیی استفاده از علفکش پاراکوات برای مدیریت علفهایهرز لوبیا (Phaseolus vulgaris L.) در قالب رهیافت تهیه بستر کاشت زودهنگام
کارآیی علفکش پاراکوات در کنترل علفهایهرز لوبیا در چارچوب رهیافت بستر کاشت زودهنگام، بهصورت کرتهای خُردشده در قالب طرح بلوکهای کامل تصادفی با سه تکرار در استان لرستان طی دو سال 1388 و 1389 مورد ارزیابی قرار گرفت. مقدار بذر لوبیا (در سه سطح 100، 150 و 200کیلوگرم در هکتار) به کرتهای اصلی و مدیریت علفهایهرز در هفت سطح (تریفلورالین، پاراکوات، بنتازون+اسیفلورفن، تریفلورالین+بنتازون+اسیفلورفن، پاراکوات+بنتازون+اسیفلورفن، وجین دستی و شاهد بدون کنترل) به کرتهای فرعی اختصاص داده شد. در سال اول آزمایش، در بین تیمارهای علفکش کمترین سطح تراکم علفهرز به تیمار کاربرد پیشرویشی علفکش پاراکوات بهعلاوه کاربرد پسرویشی علفکش بنتازون+اسیفلورفن اختصاص داشت که در مقایسه با شاهد بدون کنترل، سبب کاهش 68درصد جمعیت علفهایهرز شد. تیمارهای کاربرد پیشکاشت تریفلورالین بهعلاوه کاربرد پسرویشی بنتازون+اسیفلورفن، کاربرد پیشرویشی پاراکوات بهعلاوه کاربرد پسرویشی بنتازون+اسیفلورفن و تیمار کاربرد تنهای علفکش بنتازون+اسیفلورفن بهترتیب سبب کاهش83، 82 و 70درصد زیستتوده علفهایهرز شدند. بر اساس نتایج سال دوم آزمایش، همه تیمارهای مدیریت در مقایسه با شاهد بدون کنترل موجبات کاهش معنیدار تراکم علفهایهرز را فراهم آوردند. تیمارهای مبتنی بر کاربرد تریفلورالین با کاهش 77درصد تراکم علفهایهرز، برترین تیمارهای علفکش از نظر کنترل علفهایهرز بودند. کاربرد تلفیقی پاراکوات و بنتازون+اسیفلورفن نیز با کاهش 54درصد تراکم علفهایهرز با تیمار کاربرد تلفیقی تریفلورالین و بنتازون+اسیفلورفن تفاوت معنیداری نداشت.

ارزیابی تأثیر نوع رقم و زمان خیساندن و پخت بر ویژگی های بافت، جذب آب و میزان ترکیدگی لوبیاقرمز ایرانی
لوبیاها منبع غنی از فیبرهای رژیمی و ترکیبات شیمیایی گیاهی از قبیل فلاونوئید و فنولیک هستند و ویژگیهای فراسودمند بسیاری نشان دادهاند. در این مطالعه اثر رقم، زمان خیساندن و پخت بر جذب آب، ترکیدگی و بافت لوبیاقرمز ایرانی مورد بررسی قرار گرفت تا بهترین رقم و بهترین زمان خیساندن تعیین گردد. رقم D81083، اختر و KS31164 پس از 24ساعت خیساندن بهترتیب بیشترین میزان جذب آب را نشان دادند، درحالیکه رقم های ازنا، گلی و ناز کمترین جذب آب را داشتند (05/0P

بهمنظور بررسی تأثیر تنش خشکی و روش کاشت بر عملکرد و اجزای عملکرد دو رقم نخود، آزمایشی در مزرعه تحقیقاتی دانشکده کشاورزی دانشگاه شیراز بهصورت اسپلیتفاکتوریل در قالب طرح بلوک کامل تصادفی با چهار تکرار انجام شد. فاکتور اصلی، قطع آبیاری بر اساس مراحل رشدی نخود (دو هفته پس از سبزشدنI1=، گلدهی I2=و آبیاری نرمالI3=) و فاکتور فرعی شامل ترکیبی از دو رقم نخود (آرمان و آزاد) و روش کاشت (کرتی، درون جوی و روی پشته) بود. نتایج نشان داد که بیشترین و کمترین میزان غلاف در بوته، دانه در بوته، درصد غلاف دودانه ای، وزن100دانه، عملکرد دانه، و عملکرد بیولوژیک بهترتیب در تیمار I3 و I1 بهدست آمد و در مورد درصد غلاف پوک، روند برعکس بود. از نظر روش کاشت، بیشترین میزان غلاف در بوته، دانه در بوته، درصد غلاف دودانه ای، وزن100دانه و عملکرد دانه، در روش کاشت روی پشته بهدست آمد. بهطور کلی رقم آزاد بهطور معنیداری از عملکرد دانه بیشتری نسبت به رقم آرمان (164 در مقابل117گرم در مترمربع) برخوردار بود و رقم مناسب برای این منطقه می باشد. هرچند تحت شرایط تنش خشکی، عملکرد دانه در هر دو رقم نخود کاهش یافت، ولی تغییر در روش کاشت از کرتی به کشت روی پشته در تیمارهای قطع زودهنگام آبیاری(I1) و یا در مرحله گلدهی(I2)، بهعنوان یک روش زراعی می تواند باعث کاهش خسارت ایجادشده بهترتیب تا میزان 26 و 32درصد شود.

واکنشهای مورفولوژیک، فیزیولوژیک و بیوشیمیایی ژنوتیپهای متحمل و حساس به خشکی نخود (Cicer arietinum L.) در شرایط تنش خشکی در مزرعه
آزمایشی با هدف بررسی واکنشهای مورفولوژیکی، فیزیولوژیکی و بیوشیمیایی ژنوتیپهای حساس و متحمل به خشکی نخود در شرایط تنش خشکی با 14ژنوتیپ، شامل دو رقم تجاری جم (361MCC) و کرج( 358MCC)، سه ژنوتیپ کابلی و سه ژنوتیپ دسی متحمل به خشکی بهترتیب شامل 392MCC، 537MCC، 696MCC و 873MCC، 870MCC،10MCC و همچنین سه ژنوتیپ کابلی و سه ژنوتیپ دسی حساس به خشکی بهترتیب شامل 759MCC، 588MCC، 774MC و 39MCC، 45MCC و 101MCC در مزرعه تحقیقاتی دانشکده کشاورزی دانشگاه فردوسی مشهد در شرایط دیم انجام شد. ژنوتیپهای فوق در مرحله گلدهی، از نظر صفات وزنخشک اندام هوایی، طول ساقه، مقدار نسبی آب برگ، شاخص پایداری غشاء، پتانسیل آب برگ، سطح برگ، کارآیی فتوسیستمΙΙ (Fv/Fm)، میزان پروتئین کل محلول برگی، پرولین و میزان فعالیت آنزیم کاتالاز، مورد مقایسه قرار گرفتند. نتایج نشان داد که همبستگی مثبت و معنیداری بین میزان پرولین و پروتئین کل محلول برگی وجود دارد. وزنخشک اندام هوایی و سطح برگ نیز همبستگی مثبت و معنیداری با میزان فعالیت آنزیم کاتالاز نشان دادند. فعالیت آنزیم کاتالاز، مقدار پرولین، پتانسیل آب برگ و کارآیی فتوسیستمΙΙ در ژنوتیپهای حساس به خشکی کمتر از سایر ژنوتیپهای مورد بررسی و بهویژه ژنوتیپهای متحمل به خشکی بود که بهنظر میرسد میتوان از این صفات بهعنوان شاخصهای مناسبی جهت ارزیابی میزان تحمل به خشکی ژنوتیپهای نخود استفاده کرد. برخلاف رقم تجاری جم (361MCC)، در رقم تجاری کرج (358MCC) مقادیر بالایی از فعالیت آنزیم کاتالاز، مقدار پرولین، پتانسیل آب برگ و کارآیی فتوسیستمΙΙ مشاهده شد که این نتایج نشان از تحمل بالاتر این رقم به تنش خشکی دارد.

بررسی تأثیر تنش خشکی بر صفات مورفوفیزیولوژیک و فلوئورسانس کلروفیل گیاهچه های لوبیا قرمز (Phaseolus vulgaris L.)

(درخشان و گلی) در چهار تیمار تنش خشکی شامل 25، 50، 75 و شاهد100درصد ظرفیت زراعی بهصورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار انجام شد. نتایج نشان داد که صفات مورفولوژیک مورد بررسی از قبیل ارتفاع گیاه، سطح برگ، سطح ریشه، مجموع طول ریشه ها و نیز وزنخشک اندام هوایی و ریشه در شرایط تنش خشکی بهطور معنی داری تحت تأثیر تنش خشکی کاهش یافت (p≤0.01). همچنین نتایج نشان داد که صفات فیزیولوژیک بررسیشده از قبیل محتوای آب نسبی برگ (RWC)، شاخص پایداری غشاء (MSI)، میزان کارآیی فتوسیستمII (Fv/Fm) و عملکرد کوانتومیفتوسنتز (Y) بهطور معنی داری تحت تأثیر تنش خشکی کاهش یافتند (p≤0.01)؛ ولی مقاومت روزنه ای و شاخص کلروفیل (SPAD) در گیاهچه های لوبیا بهطور معنی داری افزایش یافت (p≤0.01). میزان افزایش نسبت ریشه به اندام هوایی رقم درخشان در تنش شدید (25درصد ظرفیت زراعی) نسبت به شاهد بیشتر از گلی بود. مقادیر RWC، MSI و Fv/Fm در تنش شدید نسبت به شاهد در رقم گلی کاهش بیشتری از درخشان نشان داد. نتایج این بررسی نشان داد که رقم درخشان احتمالاً از طریق بکارگیری مکانیسم های مختلف تحمل به خشکی نسبت به رقم گلی تا حدی کمتر تحت تأثیر تنش خشکی قرار گرفت. اغلب صفات فیزیولوژیک رقم درخشان، در سطح خشکی 75درصد ظرفیت زراعی و شاهد تفاوت معنی داری نداشتند.

روند تغییرات صفات مورفوفیزیولوژیک ژنوتیپهای نخود (Cicer arietinum L.) در پاسخ به تنش شوری ناشی از کلریدسدیم
شوری یکی از مهمترین تنشهای غیرزیستی درحالگسترش بوده که علاوه بر اثرات سمی و تغذیهای، توانایی گیاه برای جذب آب را کاهش میدهد. در مواجهه با تنش شوری، مجموعهای از واکنشهای مورفوفیزیولوژیک در گیاهان ایجاد میشود. عملکرد نخود بهدلیل حساسیت بالا به این تنش و بهدنبال برخی پاسخهای قابلتوجه در خصوصیات فیزیولوژیک و مورفولوژیک، بهشدت کاهش مییابد. در این تحقیق، پیامدهای غلظتهای مختلف تنش شوری شامل صفر، 8 و
dS.m-1 12 بر ویژگیهای مورفوفیزیولوژیک ژنوتیپهای حساس و متحمل نخود در مراحل اولیه رشدونمو گیاه بهصورت آزمایش فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار و چهار نمونهبرداری با فاصله زمانی یک هفته مورد بررسی قرار گرفت. بر اساس دادههای حاصل، با افزایش غلظت کلریدسدیم و گذشت زمان (هفته چهارم)، ژنوتیپهای MCC760 و MCC806 بهترتیب از کمترین و بیشترین آسیب بر اساس شاخصهای وزنخشک ریشه به وزنخشک اندام هوایی
(8/0 و 2برابر کاهش) و وزنخشک ریشه (7/1 و 4برابر کاهش) برخوردار بودند. در شدت بالای تنش (dS.m-1 12)، ژنوتیپ MCC760 نهتنها مقدار رنگدانههای فتوسنتزی (شامل کلروفیلهای a، b و کارتنوئیدها) بیشتری داشت، بلکه شدت کاهش رنگدانههای این ژنوتیپ، کمتر و در مقابل، ضریب پایداری کلروفیل آن (75درصد) بهصورت معنیداری بیشتر از سایر ژنوتیپها بود، بهطوریکه تفاوت آن با ژنوتیپ MCC806 بیش از 30درصد بود. درصد رطوبت نسبی در همه ژنوتیپها کاهش یافت و در هفته سوم، این دو ژنوتیپ بهترتیب با کاهش 8/1 و 4/3برابر نسبت به شاهد، کمترین و بیشترین تغییرات را نشان دادند. بنابراین در بین ژنوتیپهای مورد مطالعه ژنوتیپ MCC760 بهعنوان متحملترین ژنوتیپ به تنش شوری معرفی شد و زمان مناسب جهت ارزیابی تحمل به این صفت بر مبنای خصوصیات مورفولوژیک، هفته چهارم و بر مبنای صفات فیزیولوژیک، هفته سوم تعیین شد.

آفت پیلهخوار یکی از عوامل اصلی کاهش عملکرد نخود محسوب میشود و انتقال ژن با هدف افزایش مقاومت به این آفت، از اهداف اصلاحی در این گیاه زراعی میباشد. یکی از راهبردهای مؤثر برای تولید نخود تراریخته مقاوم به آفت پیلهخوار، استفاده از سموم طبیعی Cry از باکتری باسیلوس تورینجینسیس است. این سموم قادرند در معده حشرات فعال شده و سیستم گوارشی حشره را مختل نمایند. در مطالعه حاضر بررسی ثبات حضور ژن cry1Ac و بیان آن در نسل های سوم (T3) و چهارم (T4) حاصل از نسل دوم نخود تراریخته با ژن cry1Ac و ژن گزینشگر nptII انجام شد تا بتوان به لاینی دست یافت که تنها حاوی ژن cry1Ac بوده و ژن nptII بر اثر نوترکیبی بین دو T-DNA از آن تفکیک شود. آزمون PCR از بین 25نمونه مشکوک در نسل سوم به وجود ژن cry1Ac، در 6مورد باند مربوط به ژن cry1Ac و باند مربوط به ژن nptII را در تمامی نمونه ها نشان داد، ولی از بین این 6نمونه، در آزمون RT-PCR تنها در 5مورد ژن cry1Ac در سطح RNA بیان شد. نتایج PCR در طی نسل چهارم حاکی از وجود باند مربوط به ژن cry1Ac در 73مورد و باند مربوط به ژن nptII در81نمونه از 94مورد بود. در10نمونه از گیاهان تراریخته که حاوی ژن cry1Ac بودند، باند مربوط به ژن nptII مشاهده نگردید که این نشان دهنده آن بود که ژن cry1Ac از ژن nptII تفکیک شده است. نتایج آزمون RT-PCR نیز نشان داد که ژن موردنظر در همه گیاهان تراریخته، در سطح RNA بیان شده است. همچنین نتایج آزمون الایزا نشان داد که در تمام نمونه های مورد آزمون، پروتئین Cry1Ac در لاین های مختلف در غلظت های متفاوتی بیان شده است. در آزمون زیستسنجی، لاروهایی که با برگ گیاهان غیرتراریخته تغذیه شدند، همگی زنده ماندند، اما در مقابل لاروهایی که با برگ گیاهان تراریخته نسل T4 تغذیه شدند، همگی بهطور کامل از بین رفتند که این امر نشاندهندة بروز موفقیت آمیز فنوتیپ مورد انتظار می باشد.

بررسی مقاومت ژنوتیپهای باقلا به بیماری لکهشکلاتی (Botrytis fabae Sard.) در منطقه گرگان
بیماریهای قارچی نظیر بیماری لکهشکلاتی (Botrytis fabae sard)یکی از مهمترین بیماریهای گیاه باقلا (Vicia faba L.) در مناطق مرطوب میباشد که منجر به کاهش عملکرد باقلا میشود. این تحقیق با هدف بررسی مقاومت مزرعهای ژنوتیپهای باقلا شامل 35لاین به همراه رقم شاهد حساس ILB 365 و شاهد محلی (برکت) در برابر بیماری لکهشکلاتی در ایستگاه تحقیقات کشاورزی گرگان انجام شد. نتایج تجزیه واریانس حاکی از اختلاف ژنوتیپها از نظر عملکرد، اجزای عملکرد و صفات مرتبط با مقاومت به بیماری در سطح احتمال یکدرصد بود. بالاترین عملکرد دانه، ارتفاع بوته و وزن100دانه به رقم برکت تعلق داشت و ژنوتیپهای FLIP03-35FB، FLIP03-58FB و
FLIP03-33FB در رتبههای بعد قرار گرفتند. بهمنظور بررسی واکنش ارقام نسبت به لکهشکلاتی، میزان وقوع و شدت بیماری در فاصلههای زمانی مشخص از ظهور علایم پیشرفت بیماری تا زمان برداشت اندازهگیری و بر این اساس، منحنی پیشرفت بیماری رسم و با استفاده از آن، متغیرهای AUDPC، rAUDPC و sAUDPC محاسبه شدند. با توجه به نتایج حاصل از تجزیه واریانس، مقایسه میانگین و تجزیه کلاستر بر اساس مقادیر AUDPC وrAUDPC ، تفاوت معنیداری بین ژنوتیپهای مورد بررسی و عکسالعمل آنها نسبت به بیماری وجود داشت. ژنوتیپهای ILB 365،
FLIP03-57FB، FLIP03-43FB و FLIP03-59FB حساس و ژنوتیپهای Icarus، FLIP03-29FB،
FLIP03-30FB، FLIP03-34FB و FLIP03-52FB با دارابودن کمترین مقادیر AUDPC مقاوم تشخیص داده شدند.

بررسی الگوی بیان ژن های کیتیناز و بتا-1و3 گلوکاناز در گیاه نخود آلوده به بیماری برقزدگی
القای مقاومت نسبت به پاتوژنها از راهکارهایی است که گیاهان برای مقابله با تنشهای زیستی بهکار میبرند. آنزیمهای کیتیناز و بتا-1و3 گلوکاناز جزء پروتئینهای قابلالقاء هستند که در پاسخ به عوامل بیماریزا توسط گیاه، سنتز میشوند. در این تحقیق، بیان ژن ایزوفرمهای اسیدی و بازی این دو آنزیم در سطح مولکولی در گیاه نخود زراعی مورد مطالعه قرار گرفت. بهاینترتیب که سطح تظاهر ژنهای مذکور در نمونههای تیمارشده و شاهد، در ژنوتیپ نخود حساسMCC403 و ژنوتیپ مقاوم MCC496 نسبت به آلودگی با نژاد شِش قارچ برقزدگی (Ascochyta rabiei) در 0،6، 12، 24، 48 و 72ساعت بعد از آلودگی با استفاده ازروش ارزیابی نیمه کمیRT-PCR بررسی شد. نتایج، حاکی از افزایش بیان ایزوفرم اسیدی هر دو ژن کیتیناز و بتا-1و3 گلوکاناز پس از آلودگی بود. هردو ژن مذکور در 24ساعت اولیه پس از تلقیح، حداکثر بیان را در ژنوتیپ مقاوم نشان دادند. این در حالی است که در ژنوتیپ حساس در ساعات اولیه بعد از آلودگی، تظاهر دو ایزوفرم اسیدی به حداکثر میرسد و تا 72ساعت بعد از آلودگی، سطح بیان در هردو ژنوتیپ به کمترین سطح تظاهر میرسد. ایزوفرم بازی کیتیناز در ژنوتیپ حساس بهمیزان بیشتری بیان شد و ایزوفرم بازی
بتا-1و3گلوکاناز تظاهر پیدا نکرد. در کل، میتوان نتیجه گرفت که احتمالاً ایزوفرمهای اسیدی ژنهای کیتیناز و گلوکاناز در اعطای مقاومت در برابر بیماری برقزدگی نخود نقش دارند.

بررسی توزیع ژنهای مقاوم به ویروس موزائیک معمولی لوبیا در ژنوتیپهای لوبیا (Phaseolus vulgaris L.) با استفاده از نشانگرهای مولکولی اسکار
بهمنظور تعیین پتانسیل ژنتیکی مقاومت برخی از ژنوتیپهای لوبیا نسبت به ویروس موزائیک معمولی لوبیا، دو آزمایش مجزا هر یک در قالب طرح بلوکهای کامل تصادفی با 25تیمار (ژنوتیپهای لوبیا) و سه تکرار در دانشگاه آزاد اسلامی واحد شیراز اجرا گردید. آزمایش اول در شرایط نرمال و آزمایش دوم در شرایط آلودگی با ویروس موزائیک معمولی لوبیا انجام شد. آلودهسازی مزرعه در دو مرحله (برای اطمینان بیشتر) ظهور برگهای کوتیلدونی و ظهور اولین سهبرگچه بهصورت مکانیکی و با پودر کارباراندوم انجام گردید.پس از 21روز از آلودگی اول، جهت انجام آزمون الایزا از بوتههای هر کرت که دارای علایم ظاهری بودند، نمونهگیری شد. جهت تعیین آلودگی از روش PTA- ELISA استفاده گردید. براساس نتایج بهدستآمده، ژنوتیپهایی که دارای ژن I و حداقل یکی از ژنهای bc-3 و bc-12 بودند، بهطور معنیداری (P-value=0.001) نسبت به سایر ژنوتیپها افت عملکرد کمتری را در شرایط آلودگی نشان دادند. کمترین میزان تغییرات عملکرد مربوط به ژنوتیپ WA8528-9 (67/5درصد) بود که حاوی ژنهای I و bc-3 بود. در بین کل ژنوتیپها، ژنوتیپ11805 تنها نمونهای بود که هر سه ژن I ، bc-3 و bc-12 را داشت. میزان تغییرات عملکرد این ژنوتیپ نیز نسبتاً کم (05/9درصد) بود. درکل،60درصد ژنوتیپها حاوی ژن I ، 44درصد دارای ژن bc-3 و 8درصد حاوی ژن مغلوب bc-12 بودند. از دیگر نتایج مهم این تحقیق، واکنش منفی دو ژنوتیپ Local Khomein و Capsoli در آزمون الایزا نسبت به ویروس موزائیک معمولی لوبیا و همچنین تغییرات عملکرد کمتر از میانگین کل این ژنوتیپها بود. از آنجاییکه این دو ژنوتیپ فاقد هر سه ژن مقاوم مورد بررسی بودند، لذا این احتمال وجود دارد که ترکیبات ژنی مقاوم جدید در ژنوتیپهای Local Khomein و Capsoli وجود داشته باشد. بنابراین باتوجه به نتایج بهدستآمده از این تحقیق پیشنهاد میشود در مطالعات اصلاحی آتی در خصوص ارقام مقاوم به ویروس موزائیک معمولی لوبیا به این دو ژنوتیپ اخیر توجه ویژهای بشود.
